I turn the “AF Start” on and off on the shutter button fairly often. If I’m doing focus stacking, I want “AF Start” off but otherwise, I often leave it on. I still have one or two buttons on the back set for focus (in various modes). As far as I know, changing this setting can only be done via the menu. There is no one button set up or quick control item that is for this. And, as far as I know, the only way to trigger the shutter is via the shutter button.
The other adjustment I do fairly often is going from single shot to one of the continuous shooting modes. I know there is a one button method to switch between continuous shooting modes (from low to high or high to low). But no way to switch to or from single shot mode. In this case, it can be done via the quick controls which is nice but I wish there was a way to set up a single button for this. I’m considering setting my low continuous shooting rate to be really low like 2 frames per second and just use it as a single shot and remember to release the shutter within a half second.

You could use a custom shooting mode, C1 / C2 / C3 and have that with the alternate setup.

My aging brain follows the KISS rule. I set [AF-ON] button to AF-OFF. I don’t bother with the button programming that makes dramatic changes to camera behavior. I will use a custom shooting mode for that.

I really need to figure out how to get custom shooting modes into my work flow. I shoot all kinds of different subjects and methods but I still think I could incorporate them somehow. I just need to figure out how... (I don't mean technically but how to use them effectively.)
