is Canon pro 1000 a CMYK or RGB printer?

It uses CYMK ink but the data sent to the printer needs to be RGB.

I do not understand what you are calibrating. Do you mean you are creating custom ICC profiles?

Yes

then I suggest if you have questions about that you should be contacting the site for the company that makes the calibration device. I have never performed that task.
