My 50D all of a sudden stopped detecting a memory card

What brand and type card are you currently using in the camera? Have you tried a different card?

If the card currently has any stored images I would attempt to get them off and then do an in camera low level format and retest.
Also, it is rare but sometimes the pins in the CF memory card socket can be damaged (bent usually). Get a very bright flashlight, remove the memory card and look carefully at the bottom of the socket and examine the pins.
