how do you set the aperture in the 300mm f4 l lens?

There is no aperture ring on the lens. You set aperture with the camera body.

You can do that in either Manual (M) mode, or in Aperture Priority (AV) mode on the dial. I suggest you start wih AV Mode, because there the camera will set you a correct shutter speed for good exposure to fit your selected aperture value.

What camera?  Different cameras work, well ah, differently!  You have the white EF 300mm f4L IS lens?
