EOS 90D C. Fn II Autofocus for single shot

I use Canon 90D and set one shot as mode and single point as AF.
Do I need to update customer function settings in auto focus. (C. Fn II)?

The default custom function settings are suitable for many different photography needs - that is why they are the defaults. Therefore if you don't need to change them, then don't.

C.Fn II: 1 through to 5, and 11 only apply when using AI Servo AF, so not relevant for you

C.Fn II: 6 is mostly useful for super telephoto lenses tracking moving subjects with AI servo AF, so not really relevant.

C.Fn II: 7 & 8 - are used to limit the selectable AF methods in viewfinder and live view shooting. This can make it faster to select from the different AF point groupings.

C.Fn II: 9 determines which buttons are used to change the AF method

C.Fn II: 10 allows you to have different AF points automatically active when you turn the camera from landscape orientation to vertical. You might want a central position for your single point in landscape orientation, but prefer to have the single point nearer the top of the frame in vertical. This custom function allows this.

C.Fn II: 12 - has no effect when using single AF point.

C.Fn II: 13 decides how the AF point moves if you reach the edge of the AF area, does it stop, or wrap back to the opposite side.

C.Fn II: 14 determines how the AF points display

C.Fn II: 15 determines if the AF points will be illuminated or just black frames

C.Fn II: 16 is for lens micro adjustment. It should not be necessary for most lenses to use this setting, but some independent lenses can benefit from appropriate settings.

I suggest you study the instruction manual for your camera - particularly the section covering the AF custom functions.
